The total synthesis of compounds with interesting physical or biological properties is at the core of organic chemistry. Since only a small number of such compounds originate from natural sources, devising de novo routes to unique and artificial scaffolds requires the skills of planning and executing multi-step syntheses and having a toolbox of methodologies at one’s disposal. Often, even the best plans require modifications, fine-tuning or entire rerouting, and chemists are faced with the daily challenge of choosing the right combination of reagents and building blocks for a whole set of problems: Which building blocks are optimal? In which sequence should they be assembled? Which reactions accomplish the task best?

This text focuses on these basic considerations that govern the planning and execution of a synthesis. The author details synthetic strategies, provides criteria for ranking different synthesis plans, and provides guidelines for optimizing syntheses. References to the original work provide the reader with a means of further study of the details and original considerations. The text takes a more in-depth look at synthesis than do standard organic chemistry textbooks and is a useful complement to other books on the market.

Syntheseplanung setzt ein ungeheures Grundwissen der Organischen Chemie und eine Vielzahl praktischer Erfahrung voraus. Dieses Wissen und die Erfahrungswerte münden in spezifischen Syntheserouten für ein Zielmolekül: Retrosynthese des gewünschten Produktes führt hierbei zu einem Grundplan einzelner Bausteine, welche mit einem möglichst geringen Aufwand selektiv zum Zielmolekül zusammengesetzt werden. Details wie Schutzgruppenchemie, Anzahl Syntheseschritte und Eduktkosten stellen in der Evaluation geeigneter Strategien weitere Hürden dar.

Reinhard W. Hoffmanns Buch gibt dem Studenten das Rüstzeug für erfolgreiche eigene Syntheseplanung in die Hand. Es ist kein Lehrbuch, das Grundlagen oder fortgeschrittenes Wissen der Organischen Chemie vermittelt, sondern hilft, dieses Wissen in der Syntheseplanung praktisch anzuwenden. Das gelingt dem Autor mit leichtverständlicher Sprache und einfachen Graphiken, sowie ideal gewählten Beispielen hervorragend. "Elemente der Syntheseplanung" zeigt Taktiken auf und gibt dem fortgeschrittenen Studenten einige wertvolle Hinweise und neue Sichtweisen - gerade zur Retrosynthese, die in den Lehrbüchern zur Organischen Chemie vernachlässigt wird. Hoffmann gelingt es an einigen Stellen, Wissen zu aktivieren. Studenten, welche in den Kapiteln zu wichtigen Synthons aktiv mitarbeiten, werden einen grossen Nutzen durch die zusätzliche Sichtweise aus Hoffmanns Buch erfahren.

Für fortgeschrittene Studenten und Leser, welche sich für Syntheseplanung interessieren, ist die Lektüre von "Elemente der Syntheseplanung" höchst empfehlenswert!
